Clinical impact and prognostic value of CD147 and MMP-7 expression in patients with pancreatic ductal adenocarcinoma

چکیده
Aim: Pancreatic ductal adenocarcinoma (PDAC) is a lethal disease with short overall survival. It is of great significance to identify new biomarkers to predict prognosis of PDAC patients after surgery. Methods: In this study, we examined the expression of CD147 and MMP-7 in 90 PDAC neoplastic tissues and paired non-neoplastic tissues from patients who underwent surgery and followed-up for as long as 87 months by immunohistochemistry staining. Χ2 test, Kaplan-Meier and log-rank test were used to analysis the correlation between CD147, MMP-7 expression and clinicopathological features. Prognostic analysis was carried out by using multivariate Cox regression models. Results: Our results showed that expression of both CD147 and MMP-7 were increased in neoplastic tissue (P<0.001). MMP-7 expression in neoplastic tissue positively correlated with lymph node metastasis (P<0.05). Both CD147 and MMP-7 expression in neoplastic tissue had significantly impact on overall survival in patients with PDAC (P<0.05). More importantly, MMP-7 expression was correlated with the expression of CD147 in PDAC patients. Of 81 analyzed specimen, concurrent expression of CD147 and MMP-7 was found in 29 patients and had significantly negative impact on overall survival. Multivariate analysis determined that co-expression of CD147 and MMP-7 was an independent factor for prognosis. Conclusion: Our results indicate that CD147 and MMP-7 expression is associated with overall survival in PDAC patients and concurrent expression of CD147 and MMP-7 could better predict the prognosis after surgery. The conclusion of this study can help physicians to make better follow-up decision to monitor the disease progression in PDAC patents.

Comprehensive analysis of matrix metalloproteinase and tissue inhibitor expression in pancreatic cancer: increased expression of matrix metalloproteinase-7 predicts poor survival.
PURPOSE To enable the design of improved inhibitors of matrix metalloproteinases (MMPs) for the treatment of pancreatic cancer, the expression profiles of a range of MMPs and tissue inhibitors of MMPs (TIMPs) were determined. EXPERIMENTAL DESIGN Nine MMPs (MMPs 1-3, 7-9, 11, 12, and 14) and three TIMPs (TIMPs 1-3) were examined in up to 75 pancreatic ductal adenocarcinomas and 10 normal pancr...
متن کاملExpressions of Matrix Metalloproteinases 2, 7, and 9 in Carcinogenesis of Pancreatic Ductal Adenocarcinoma
Pancreatic ductal adenocarcinoma (PDAC) is a highly fatal disease, usually diagnosed in an advanced stage which gives a slight chance of recovery. Matrix metalloproteinases (MMPs) are a family of zinc-dependent endopeptidases that participate in tissue remodeling and stimulate neovascularization and inflammatory response. The aim of the study was to evaluate the expression of MMP-2, MMP-7, and ...
متن کاملPotential prognostic biomarkers in pancreatic juice of resectable pancreatic ductal adenocarcinoma
Despite potentially curative surgery pancreatic cancer has a dismal prognosis. Serum cancer antigen 19-9 (CA 19-9) correlates with tumor burden, resectability and survival in patients with pancreatic ductal adenocarcinoma. Identification of novel biomarkers may facilitate early diagnosis of pancreatic cancer and improve survival. Pancreatic juice is a rich source of cancer-specific proteins ren...

MicroRNA-21 (miR-21) was reported to be overexpressed and contributes to invasion and gemcitabine resistance in pancreatic ductal adenocarcinoma (PDAC). The aim of this study was to evaluate whether miR-21 expression was associated with the overall survival (OS) of PDAC patients treated with gemcitabine and to provide mechanistic insights for new therapeutic targets. miR-21 expression was evalu...
متن کاملPrognostic Value of HLA-G in Malignant Liver and Pancreas Lesions
Background: Human leukocyte antigen (HLA)-G is a nonclassical HLA class I molecule with modulatory effects on NK and T cells. Because HLA-G expression is frequently detected in different solid tumors, it may be involved in tumor immune evasion. Objective: This study was designed to elucidate the prognostic value of HLA-G in hepatocellular carcinoma (HCC) and pancreatic adenocarcinoma (PADC). Th...
